Job Summary
Conduct high-throughput expression of novel multi-specific biologics in CHO cells, optimizing chain ratios and vector strategies for batch and fed-batch bioreactors. Generate stable cell lines with enhanced titers and monomer purity using targeted integration technologies like FACS-based single-cell sorting and ddPCR. Troubleshoot challenges involving glycosylation, aggregation, and chain pairing while implementing automation platforms to streamline workflows. Purify and characterize analytical samples via HPLC, then independently design, execute, and rigorously report experimental data to support drug discovery projects.
Required Qualifications
- Bachelors or Master's in biology, chemistry, biochemistry or related field
- 5+ years of experience for BS degree holders
- 2+ years of experience for MS degree holders
- Hands-on experience with cell culture and expression of monoclonal antibodies, bi-specifics and multi-specifics in CHO cells lines including thaw, culture, expression and cryopreservation
- Prior experience in generation of stable CHO cell lines expressing complex multi-specific molecules
- Prior experience in process development, process optimization, and automation
- Experience with analytical characterization of multi-specifics
- Experience with targeted integration, FACS, and ddPCR
- Demonstrated proficiency in vector design
- Prior experience in mammalian expression using Wave bioreactors
- Demonstrated proficiency in overcoming problems (i.e. poor chain pairing, low expression, aggregation, glycosylation) with challenging therapeutic multi-specific proteins
