Job Summary
Develop and deploy assays to systematically evaluate fixation quality, protein retention, and antigenicity across tissue preparation conditions. Establish quantitative metrics for molecular readout and drive improvements in high-quality morphology and protein readouts by collaborating with molecular biology, imaging, and computational teams. Work closely with in vivo research technicians to design experiments, evaluate results, and optimize standard operating procedures. Generate in-house pipelines for binder design, discovery, and validation of epitope-binder pairs in hydrogels to improve barcode readout of synaptic proteins. Develop novel highly multiplexed protein readout strategies for molecular annotation of synapses and cell types. This role reports to the Barcoding Lead and focuses on advancing whole-brain connectomics through engineering approaches to the full pipeline from sample preparation to molecular readout.
Required Qualifications
- equally at home at the bench, the microscope, and in quantitative data analysis
- brings an engineering approach to the full pipeline, from sample preparation through molecular readout tools
- owns both optimizing the physical preparation of the tissue and engineering the reagent panels across two target types: comprehensive synapse identification and barcode epitopes
- define what high-quality tissue means quantitatively
- drive the improvement of high quality morphology and protein readout
- working closely with molecular biology, imaging, and computational teams to support whole-brain connectomics
